The MS medium supplemented with 5μM IAA in combination with 10μM zeatin or 10μM BA in the absence of Agrobacterium tumefaciens gave the greatest number of shoot regeneration. Shoot induction after co-cultivation with Agrobavterium tumefaciens was best achieved on a selection medium supplemented with 1.25μM IAA and 2.5μM zeatin, and occurred in 2-4 weeks. Among the three varieties tested Chunghong gave the highest regeneration frequency when co-cultivated with Agrobacterium tumefaciens on a kanamycin-containing selection medium.
