A genomic clone for potato β-1,3-glucanase was isolated to study the regulatory expression mechanism of the plant gene. A potato genomic DNA library was constructed with potato total DNA isolated from potato (Solanum tuberosum L. cv. sumi) leaves. Genomic DNA clone for β-1,3-glucanase was isolated by plaque hybridization of the library using the cDNA clone for tobacco β-1,3-glucanase as a probe. A genomic clone. λGlc1, was isolated from the library and restriction mapping was carried out. The Southern blot ananysis showed that the genomic fragment of 1.6kb harbored the β-1,3-glucanse gene of potato. The structure of the genomic clone was determined by nucleotide sequencing, which, with the result of RNA mapping with nuclease S1 together, shows that the β-1,3-glucanase gene of potato consists of 2 exons and 1 introns, 1,269bp and encodes 193 amino acids. The transcription site of β-1,3-glucanase was determined to be at 42bp upstream of translation initiation site, ATG by primer extension experiment.
