Rice chitinases are encoded by a small multigene family. To investigate the overall organization of rice chitinase genes and the regulatory mechanisms of the expression of the gene, we have isolated two different genes encoding the class Ⅰ and class Ⅱ chitinases (Rcht-A and Rcht-B) from a rice (Oryzae sativa cv. IR 36) genomic library using a partial rice chitinase cDNA clone, pRCH08, as a probe. The complete nucleotide sequences of two rice chitinases genes, Rcht-A and Rcht-B were analyzed. The Rcht-A chitinase gene contained two introns of 89 and 984 bp, and encoded a acidic chitinase of 261 amino acids with a putative 29 amino acid N-terminal signal peptide, and had the C-terminal conserved catalytic domain but did not contain the N-terminal cysteine-rich "hevein" domain, which are typical for class Ⅱ chitinases. The Rcht-A is a structurally diverse familly from other known rice chitinase genes. Rcht-A is most homologous to tomato class Ⅱ chitinase with 64.4% homology in amino acid sequence level. And the RGC-B chitinase gene contained an open reading frame with no introns, encoding a polypeptide of 320 amino acids with a isoelectric point of 4.66. This polypeptide consists of a 18 amino acid signal peptide, the N-terminal cysteine-rich domain and a conserved catalytic domain, which is characteristics of class I chitinase.
