This study was conducted for the identification of pure Landrace, Large White Duroc, Berkshire and Hampshire breeds which are mainly maintained in Korea using breed-specific DNA markers. We were used known KIT and MC1R mutations related coat color in pigs, and the particular phenomenon in the porcine mitochondrial genome such as the 11-bp duplication at D-loop region and alternative usage of initiation codon of ND2. The KIT mutation was given the result to distinguish white and colored phenotypes. Duroc breed could be assorted from other colored breeds by the MC1R mutation N121D. Discriminating Landrace and Large White was possible by the 11-bp duplication of D-loop region and alternative initiation codon of ND2. The result of blind test with every 5 animals of Landrace, Large White, Duroc, Hampshire and Berkshire showed a perfect match between estimation by tests using makers and real types of breeds. In conclusion, identification of Landrace, Large White and Duroc breeds was absolutely possible using the procedure designed in this study. The KIT gene that controls dominant white is on the pig genome with single or duplicate types. Two types of KITs are observed and classified by the point mutation (G-to-A base substitution) of the splice-donor site in the first nucleotide of intron17. The normal (G) and spliced (A) types are assigned as the KIT1 and KIT2 versions, respectively. Furthermore, KITs are classified as normal(C) or duplicate (G) copies. The two copies are distinguished by the C-to-G base substitution at the 3'-end of the duplicate or normal copy of the gene. This information allows genotypes to be defined by the ratio of KIT2 to KIT1+KIT2 and duplicate versus duplicate+normal copies of KIT. If the Landrace and the Large White may not be fixed to the Dominant white allele, colored commercial pigs could be born from the three-way cross using F1 (Landrace x Large White or vice versa) sows x Duroc (i/i) boars, which is the most common case in Korea. DNA technology and markers that simultaneously applied PCR (polymerase chain reaction)-LCR (ligation chain reaction) to survey the relative ratio of KIT1and KIT2, or duplicate and normal copies of KIT were developed in this study.
